Abstract Mycorrhiza is a mutualistic symbiosis between some plant species and diverse group of soil fungi. Mycorrhizal categorized into discrete types based on the structural differences taxonomic position involved plants The main mycorrhiza include arbuscular, ericoid, orchid, monotropoid ectomycorrhiza, latter being focus this review. Identifying ectomycorrhizal fungi (EMF) crucial for understanding ecology preservation rare endangered plants, habitats. Thus, since discovery new identification methods have been constantly developed. This review tracks history EMF methods, presents their limitations aims to discern frontier field. early were morphology anatomy EMF, which allowed ectomycorrhizae formed by different be distinguished. Recognizing that them, however, was difficult. introduction molecular analysed taxon-specific DNA barcodes breakthrough in identification, internal transcribed spacer gene adopted as routine fungal barcoding sequence. from randomly sampled roots or polymerase chain reaction (PCR)-based results taxa missed underreported. addressed bulk sampling combined with environmental metabarcoding which, turn, yield lower resolution identification.
